The invention relates to medicine, namely to a method for diagnosis of viral hepatitis B with the determination of AgHBs and its subsequent confirmation in persons with accidental exposure such as medical personnel, drug users, patients undergoing hemodialysis treatment, etc.Summary of the invention consists in that in two strips with wells is used AgHBs free human serum, human serum negative to AgHBs antibodies, serum with antibodies to AgHBs with a titer of more than 100 IU/ml and serum of the human being tested in the first strip, and in the second strip is used AgHBs free human serum, fetal bovine serum for calibration, inactivated human serum, containing calibrated AgHBs and serum of the human being tested. After incubation at a temperature of 18…24°C, for 30 min, diluted conjugate 1 : 20 of rabbit or mouse anti-species globulin and horseradish peroxidase is added in all samples, then the samples are incubated at a temperature of 37°C, for 120 min. Further, a chromogen solution containing 0.02% hydrogen peroxide solution, 4% dimethylsulfoxide solution and 0.03% tetramethylbenzidine solution is added and incubated at a temperature of 18…24°C, for 30 min, after which the optical density values are determined by the photometric method at the wavelength of 450…620 nm. |
