PROBLEM TO BE SOLVED : To provide antibodies variants having decreased or increased ability to mediate complement-dependent cytotoxicity (CDC) due to modifications at the C-terminus of their heavy chains, and to provide methods of generating such antibodies, as well as to provide nucleotide constructs and host cells suitable for the production of the antibodies. SOLUTION : A method for producing an antibody in a host cell, the antibody comprising at least a heavy chain, the method comprising the following steps : a) providing a nucleotide construct encoding the heavy chain, the construct not encoding a lysine residue at the C-terminus of the heavy chain, b) expressing the nucleotide construct in a host cell, with the proviso that the host cell is not a CHO cell or a moss cell, and c) recovering the antibody from a cell culture of the host cell.
